N-Histofine® Simple Stain AEC Solution (from Nichirei Biosciences) is a pre-made chromogen-substrate solution for peroxidase-based immunohistochemical staining. AEC (aminoethyl carbazole) produces red precipitates at target antigen sites reacting with peroxidase. For more information please visit our Nichirei Biosciences Dashboard.

Advantages
1) Ready to use solution requires no advance preparation
2) Solution stable for 18 months after manufacture if stored properly

DESCRIPTION
N-Histofine® Simple Stain AEC Solution is designed as one of reagents to visualize antigen binding primary antibody on tissue sections. I-2.

INTENDED USE
N-Histofine® Simple Stain AEC Solution is designed to allow immunohistochemical tests, to reveal antigens that reacts with a user-supplied primary on tissues and cells. The product is characterized by a coloring reagent/substrate for coloring a detection system reagent labeled with peroxidase. The product is characterized by a coloring reagent/substrate for coloring a detection system reagent labeled with peroxidase. The product is one of the specific chromogen/substrate solutions for peroxidase and develops red colored precipitate by reaction with peroxidase. The result provides a physiological or pathological state. For In-vitro Diagnostic Use. For manual staining. Staining Results are qualitative. For formalin-fixed paraffin-embedded human tissue sections. For professional users. Interpretation must be made within the context of the patient’s clinical history and other diagnostic tests by a qualified pathologist. I-3.

PRINCIPLE
The antigen / antibody / Universal Immuno-peroxidase Polymer complex can be prepared by allowing the reagent to react with a primary antibody bound to the antigen on tissue section. The enzymatic activity of this complex results in a colored deposit, thus staining the antigen site. II.

USAGE
Adjust N-Histofine® Simple Stain AEC Solution to room temperature prior to use. React the objective tissue section with peroxidase conjugate in advance. At the same time, staining of Negative control slide using PBS in place of primary antibody is recommended to be prepared for checking the staining result. For color development, incubate the tissue section on the slide with 1-2 drops of -Histofine® Simple Stain AEC Solution using Dropping bottle at room temperature for 5-20 minutes. Adjust the reaction time by microscopic observation by contrast with the Negative control slide. After adequate coloration, wash the slide with tap water. This red deposit is soluble in organic solvent. Therefore, after counterstain, the slide can be mounted with water-soluble mounting media. IV.

STAINING PROCEDURE
Refer to the instruction* of -Histofine ® Simple Stain MAX PO (MULTI) for staining procedures such as Principle, Materials required but not supplied, Specimen preparation, Control slides, Interpretation of results and General limitation. Validation is required when other detection systems besides above are used. *Download of the instruction is available at following website: http://www.nichirei.co.jp/bio/english/products/pdf/CE_MAXPO_MULTI.pdf V.

STORAGE, HANDLING & SHELF LIFE
1. Store at 2-8°C.
