Purpose of Use:
Hematoxylin-Eosin is a common staining technique used in a wide range of tissues. Through this method, nuclei can be visualized as blue, muscle in red, connective tissue in pink and nucleic acid associated with proteins in purple. Mayer’s Hematoxylin does not contain mercury.
Uses and sample preparation:
Slides should be deparaffinized through xylene or substitute and descending grades of alcohol to distilled water prior to staining. Designed for research use only, it is not approved for human or animal use, or clinical diagnosis.
Components:
- Mayer’s Hematoxylin Reagent
- Eosin Reagent
Short procedure:
- Add Mayer’s Hematoxylin
- Wash with warm tap water
- Add Glacial acetic acid
- Wash with tap water
- Add Eosin solution
- Wash with tap water
- Dehydrate with graded ethanol, clear and coverslip
Staining characteristics:
Nuclei can be visualized as blue, muscle in red, connective tissue in pink and nucleic acid associated with proteins in purple.
