Bone formation culture kit (mouse)
The osteogenic culture kit (mouse) is a cell culture kit that combines a group of cells purified from mouse bone marrow and two types of culture media. Cells proliferated in growth medium can be differentiated into osteoblasts in osteogenic medium and calcium can be deposited.
Background
Bone marrow can be divided into hematopoietic cells and supporting bone marrow stromal cells. Some of these bone marrow stromal cells also contain undifferentiated mesenchymal stem cells, which can be induced to differentiate into osteoblasts, chondrocytes, and adipocytes.
Composition
This culture kit is a combination of cells and two types of media for easy osteogenesis.
- Frozen cells 1×10 6 cells 1 tube
- 1 bottle of growth medium 125 mL
- 1 bottle of Osteogenic Medium 250 mL
Origin of cells
Cryopreservation of cells isolated from bone marrow of ICR mice
Main component of culture medium
Product name | Main ingredient |
growth medium | α-MEM, serum, others |
osteogenic medium | α-MEM, serum, dexamethasone, β-glycerophosphate, ascorbic acid, others |
Culture example
Figure 1 Cell morphology
A: The day after seeding, B: Confluent, C: Cells cultured in osteogenic medium for 3 weeks (cultured in a 35mm dish)
Cells cultured in 24-well plates according to the Figure 2
protocol and cultured in osteogenic medium were detected for alkaline phosphatase activity and calcium deposition. Alkaline phosphatase staining kit (Part number: AK20) was used to visualize alkaline phosphatase activity, and calcification staining kit (Part number: AK21) was used to stain areas with calcium deposits. I found that the deposition was progressing.
Frequently asked questions and answers (FAQ)
Q: Does the dedicated medium contain β-glycerophosphate?
A: Contains β-glycerophosphate in osteogenic medium. β-glycerophosphate is added to promote calcification.
