When the new coronavirus infects a cell, the spike protein on the surface of the virus binds to ACE2 on the cell surface. Drugs that block this binding will prevent viral infection.

Background
Angiotensin-converting enzyme 2 (ACE2), also known as ACE homolog, is an integral membrane protein that is highly homologous to ACE. ACE2 was known as one of the factors that work in the renin-angiotensin system, which is involved in blood pressure regulation. Recently, it has been revealed that when the novel coronavirus that causes COVID-19 infects human cells, it binds to ACE2 present in the cell membrane and is then taken into the cell, and ACE2 is also thought to be a receptor for the novel coronavirus. human ACE2 protein with His tag (product number: HAK-ACE2_UL-1) on a 96-well plate and adding SARS-CoV-2 spike protein RBD with rabbit Fc tag (product number: HAK-SPD_UL-1) or biotinylated SARS-CoV-2 spike protein RBD with rabbit Fc tag (product number: HAK-SPD_bio-1), which is a fusion protein in which the receptor binding domain of the viral protein is bound to rabbit Fc, and detecting with an HRP-labeled secondary antibody or streptavidin.

Usage Example
- Binding assay
- 0.156-10µg/mL of solid-phased human ACE2 protein/His tag (100µL/well) was bound to 0.8µg/mL of biotinylated SARS-CoV-2 spike protein RBD/rabbit Fc tag, and detected with HRP-labeled streptavidin. The
linear range of the calibration curve was 0.156-1.25µg/mL (Figure 2). - Competitive binding inhibitor screening
- A binding inhibitor (SARS-CoV-spike protein RBD/rabbit Fc tag) was added at 10-5000ng/mL to the binding reaction of 1µg/mL solid-phased human ACE2 protein/His tag (100µL/well) and 600ng/mL biotinylated SARS-CoV-2 spike protein RBD/rabbit Fc tag, and the reaction was detected with HRP-labeled streptavidin. The 50% inhibitory concentration (IC50) was 140ng/mL (2.7nM) (Figure 3).
