Quantification of human MSC-derived exosomes

Human mesenchymal stromal cells (hMSC) are multipotent cells with both regenerative and immunomodulatory activities making them an attractive tool for cellular therapy. Human mesenchymal stromal cells (hMSC) are multipotent cells with both regenerative and immunomodulatory activities making them an attractive tool for cellular therapy. The conventional way to quantify exosomes is indirect. For example, it measures encapsulated protein abundance or performs Nanoparticle Tracking Analysis to determine the size distribution profile of small particles. The drawback for these methods is that they require exosome purification utilizing ultracentrifugation, for instance. The direct methods to quantitate exosomes in body fluids or culture supernatant is extremely limited, and there were no common methods available until now.

This product is a two-step sandwich ELISA kit to quantitate the exosomes which are secreted from hMSC and positive in both CD63 which is a dominant tetraspanin in hMSC and CD90 which is proposed as a marker for hMSC-derived exosomes utilizing high-performance antiCD90 and anti-CD63 antibodies.

  • ●Directly quantitate HER2-positive exosomes in human blood samples or cell culture supernatant.
  • ●No special equipment is required. Standard microplate reader capable of reading at 450nm will do the job.
  • ●Utilize HER2/CD9 fusion protein (Standard Protein), instead of unstable/hard to store exosome itself, to implement stability and reproducibility.
  • ●Normalization with a standard curve using HER2/CD9 fusion protein (Standard Protein) enable to relative quantitate each sample.
  • ●Detect human HER2-positive exosomes by two-step sandwich method using immobilized anti-HER2 antibody and HRP conjugated anti-CD9 antibody.

UE6E7T-3 cells, immortalized MSC, were seeded in a 12 well dish (1 x 104 cells/cm2) with 1mL POWEREDBY10 medium. The supernatant in each well was collected day by day, then centrifuged at 2,000 x g for five minutes to remove debris. The frozen stocks of the supernatants were thawed and directly measured with this kit.
