Much of modern cell and molecular biology relies on the isolation of pure, functionally intact populations of cells, subcellular organelles and macro-molecules in high yield and both research and clinical applications provide an impetus for the development of improved separation technology.
The most commonly used method of separating suspensions of mixed populations of biological particles is centrifugation in density gradients - and the choice of medium used to form the gradients is critical for the retention of particles function. Since the early sixties Nycomed have been developing and producing iodinated compounds as X-ray imaging agents. It was recognized early on that these compounds were also well suited to the formation of density gradients and offered new opportunities for improved methods of separating biological particles, especially osmotically-sensitive cells and subcellular organelles. Nycomed has therefore been actively involved over the last 30 years in the research and development of density gradient media.

GenomONE™ HVJ-Envelope Kits for
Cell Fusion and Truly Unique Transfection.

GenomONE™ series products from Ishihara Sangyo KK (Japan) repurpose the membrane fusion ability of inactivated Sendai virus (aka Hemagglutinating Virus of Japan (HVJ) for laboratory use. GenomONE™-Si EX, GenomONE™-GX EX, and GenomeONE™-Neo EX kits are purposed specifically for transfection. GenomONE™-CF EX is purposed for cell fusion. Both kit types contain all essential reagents and buffers for use.
Inactivated, purified HVJ particles (HVJ-Envelope, HVJ-E) are non-infectious vesicles ~300 nm in diameter that retain the viral HN and F surface proteins involved in cell recognition and membrane fusion, respectively. HN protein attaches HVJ-E to sialic acid-containing cell receptors found on almost all animal cells. Binding of HN protein to the receptor induces a conformational change allowing the F protein to trigger membrane fusion.
Permeabilized HVJ-E vesicles readily incorporate molecular cargo. Treatment of cells or tissue with cargo-loaded HVJ-E vesicles results in membrane fusion and cargo delivery directly into the cell cytoplasm.
In contrast, molecular cargo introduced with conventional cationic-lipid based transfection reagents enter cells via endocytosis and may therefore be damaged by lysosomal activity.
