Glucose intracellular uptake measurement kit (wide range, fluorescence method)
Easy measurement of sugar uptake into cells by non-RI method
The enzymatic reaction by glucose-6-phosphate dehydrogenase (G6PDH) produces NADPH in proportion to the amount of 2DG6P, and the fluorescence intensity of the fluorescent substance generated from the fluorescent substrate accompanying the oxidation of NADPH is measured.
Insulin is a hormone that lowers blood sugar, and its action is known to promote sugar transport activity mainly in insulin-sensitive organs such as adipocytes and skeletal muscles, and to take sugar from extracellular to intracellular.
Glucose uptake is generally measured using radiolabeled 3 -O-methyl-D-glucose or 2-deoxyglucose (2DG) such as 3H. Since this method uses radioactive isotopes, it can only be used in RI controlled areas, and there are strict restrictions on its use, so it cannot be used at the normal laboratory level.
This kit is a kit that can easily measure the amount of sugar uptake into cells by a non-RI method that has no safety or regulatory issues.
Fig. 1 Kit measurement principle
2DG taken into the cell is phosphorylated to 2-deoxyglucose-6-phosphate (2DG6P) by hexokinase, but it remains in the cell without proceeding to the next enzymatic reaction.
