Magnetic Cell Separation Kit for
Human CD44v9+ Cancer Stem Cells
Enrichment of cancer stem cells is easily possible
The Magnetic Cell Separation Kit for Human CD44v9+ Cancer Stem Cells is a magnetic particle conjugated with a monoclonal antibody that binds with high affinity to the membrane antigen CD44v8-10, which is highly expressed on cancer stem cells. Cancer stem cells can be enriched.
Background
In recent years, it has become clear that cancer stem cells are the root cause of canceration, such as the formation of cancer tissue, its metastasis, and recurrence. Therefore, it is considered essential to eliminate cancer stem cells in order to eradicate cancer, and cancer stem cells are attracting attention as the ultimate target for cancer therapy. However, since cancer stem cells exist in an extremely small amount of less than a few percent in tissues and cell lines, it has been difficult to develop therapeutic drugs targeting them and to analyze their functions.
"Anti-CD44v antibody" is a monoclonal antibody that binds with high affinity to the membrane antigen CD44v8-10, which is highly expressed in cancer stem cells. concentration becomes easily achievable. By concentrating cancer stem cells, it is possible to construct a "sphere culture system" used as an in vitro evaluation system for cancer stem cells and a "mouse lung metastasis model" used as an in vivo evaluation system. This makes it possible to elucidate the mechanism of cancer development and metastasis, and to screen therapeutic drugs for cancer stem cells. "Anti-CD44v antibody" will be a powerful tool for the development of new therapeutic drugs targeting cancer stem cells.

Workflow of Cancer Stem Cell Enrichment Kit
Experimental example using flow cytometry
Cells stably expressing CD44v9-GFP were isolated and analyzed by flow cytometry
Fig. 1. Ratio of CD44v9-expressing cells among RV3 beads-positive cells Cell groups in which 293F cells stably expressing
CD44v9-GFP and parental cell line 293F cells were mixed at an arbitrary ratio (A, B, C). was separated using this product.
CD44v9-expressing cells were enriched from 4.97% (A), 15.3% (B), 55.7% (C) before isolation to 86.4% (A), 97.1% (B), 99.0% (C) after isolation. rice field.
Isolate cancer cells and analyze by flow cytometry

RV3 bead-positive cells RV3 bead-positive cells were stained with PE-labeled anti-rat IgG antibody and analyzed by flow cytometry. In human prostate cancer cell line PC-3 (A) and human pancreatic adenocarcinoma cell line AsPC-1 (B), the ratio of CD44v9-expressing cells also increased compared to before isolation.
