Ability to measure sugar uptake into cells without using RI
2-deoxyglucose (2DG) metabolic rate measurement kit
Glucose metabolism measurement kit (intracellular uptake activity measurement kit)
This product is an epoch-making measurement kit that measures the amount of sugar uptake after the non-radioactive substance 2-deoxyglucose is taken up into cells, and the cell extract is subjected to a two-step enzymatic reaction and enzyme cycling method. . It is possible to measure the amount of 2DG uptake into cells without using RI, and it is useful for research on metabolic syndrome including drug discovery.
Background
Glucose, an important substance in the metabolic system, is taken up into cells to generate ATP through glycolysis, and is also used in adipocytes to synthesize triglycerides, which are neutral fats. Measuring the amount of glucose uptake into cells provides information on the glucose uptake capacity and glucose consumption of the cells themselves, and is an extremely useful technique for metabolic syndrome research, including drug discovery. The measurement principle is based on the method of quantifying residual 2DG in cells using 2-deoxyglucose (2DG), which is not metabolized by glycolysis. However, the mainstream of conventional methods is the RIA method, in which 2DG is labeled with RI, and the development of a new measurement method that does not use RI has been desired.
This product is an epoch-making measurement kit that can measure the amount of 2DG uptake into cells by a colorimetric method using an enzyme without using RI. Click here for simple type (wide range, fluorescence method) products .
Features
- 2DG uptake in cultured cells can be measured.
- Since it is a non-RI method, it can be measured at the normal laboratory level.
- Since 2DG6P is measured, intracellular sugar uptake can be quantified and is not affected by extracellular 2DG.
- It can be detected with high sensitivity by the enzymatic cycling method.

Measuring principle
2DG taken into the cell is phosphorylated to 2DG6P by hexokinase, but it remains in the cell without proceeding to the next enzymatic reaction. Therefore, the cells contain endogenous G6P and 2DG6P, but after the endogenous G6P contained in the sample is degraded in the first step of this kit, it is taken up into the cells in the second step. NADPH is produced in proportion to the amount of 2DG6P obtained and detected with high sensitivity by the enzymatic cycling method.
